NSAIDs split into three classes by effect on glycosaminoglycan synthesis in OA cartilage: stimulatory (aceclofenac, tenidap, tolmetin), neutral (piroxicam, tiaprofenic acid, aspirin), and inhibitory (naproxen, ibuprofen, indomethacin, nimesulide).

Population

~650 arthritic and ~180 non-arthritic human cartilage specimens cultured over 12 years with a standardized method

Intervention

Therapeutic concentrations of individual NSAIDs applied to explants

Untreated explant cultures

Limitations

Explant culture does not establish clinical harm; single-laboratory program; drug exposure and tissue sourcing differ from in-vivo pharmacology.
