Pharmacokinetic and safety profile of trans-resveratrol in a rising multiple-dose study in healthy volunteers

Even six-times-daily dosing produced low plasma concentrations: peak levels of 3.89, 7.39, 23.1 and 63.8 ng/ml at the four doses, with trough levels at or below 10 ng/ml and inter-individual variation above 40%. Half-life was 2-5 hours on repeated dosing, and bioavailability was higher after morning administration.

Healthy adults in four dose groups of ten
Intervention
trans-Resveratrol 25, 50, 100 or 150 mg six times daily for 13 doses
Comparator
Placebo within each group
Limitations
Healthy young volunteers; free trans-resveratrol only, so the pharmacologically ambiguous conjugates are not captured. Explains why chondrocyte-experiment concentrations are unreachable by any swallowed dose.
