Resveratrol is not a direct activator of SIRT1 enzyme activity

Beher D, Wu J, Cumine S, Kim KW, Lu SC, Atangan L · Chemical Biology & Drug Design

Preclinicalcounts toward this tier

Resveratrol failed to activate SIRT1 against either a native p53 peptide or acetylated PGC-1alpha isolated from cells, in vitro or in cell-based assays; the untagged peptide was not even a substrate without its fluorophore. The authors concluded resveratrol's effects are unlikely to run through direct enzyme activation.

Population
Purified SIRT1 with p53-derived peptide and cell-derived acetylated PGC-1alpha
Intervention
Resveratrol
Comparator
Fluor de Lys tagged substrate
Limitations
In vitro; industry laboratory. Rules out one mechanism, not all of them.
