Human OA chondrocytes show reduced electron-transport-chain Complex I, II, and III activity versus normal chondrocytes, and mitochondrial dysfunction plausibly feeds oxidative stress, cytokine-driven catabolism, matrix calcification, and chondrocyte apoptosis — the rationale for testing mitochondria-targeted drugs in joints.
