Cellular senescence in osteoarthritis pathology

Osteoarthritis chondrocytes express telomere attrition, an activated DNA-damage response, reactive-oxygen-species secretion, senescence-associated beta-galactosidase, raised p16 and cytokine-driven matrix metalloproteinases relative to chondrocytes from normal cartilage. Because mitosis in normal cartilage is very low, the review attributes chondrocyte telomere shortening to oxidative stress and abnormal mechanical loading rather than to replication alone, and warns that beta-galactosidase staining in cultured cells from diseased joints is not a reliable indicator of senescence in vivo.
