Vitamin K2 and D in Patients With Aortic Valve Calcification: A Randomized Double-Blinded Clinical Trial

The largest MK-7 calcification trial is null on its primary endpoint: aortic valve calcification rose 275 units on treatment and 292 on placebo, a difference of 17 units (95% CI -86 to 53, P=0.64), with no effect on valve area, jet velocity, aortic or coronary calcification, or surgery rates — despite inactive matrix Gla protein falling sharply (-212 vs +45 pmol/L, P<0.001). Carboxylation improved and the calcium did not stop.

Community-dwelling men, mean age 71, with an aortic valve calcification score above 300 arbitrary units, 24 months

Intervention

MK-7 720 ug plus vitamin D 25 ug daily

Elderly men with established, advanced valve calcification, where any brake may come too late; MK-7 given with vitamin D, so the two cannot be separated; 24 months; registered as NCT03243890.
