Traces the one link this entry's joint chain was missing a modern statement of: SIRT6 is NAD+-dependent, its activity falls as NAD+ falls with age, and in chondrocytes it regulates mitochondrial number and membrane integrity, limits reactive oxygen species and supports mitochondrial DNA transcription and biogenesis. Supplementing NAD+ precursors is presented as one route to raising it, alongside specific SIRT6 activators. The review is candid about where that stops: SIRT6 targeting has not been verified in humans, no systematic clinical trial has evaluated the efficacy or safety of a SIRT6 activator, and SIRT6 has not been validated as a biomarker.
