The field's most complete audit. Rodent studies frequently showed metabolic, mitochondrial and functional gains. In humans, oral NR and NMN consistently hit their biochemical target — circulating and cellular NAD+ rose — and were well tolerated over weeks to months, but effects on functional, metabolic, vascular and other healthspan outcomes were heterogeneous and often null or endpoint-specific. No outcomes trial of intravenous NAD+ for anti-ageing exists.

- Narrative synthesis without pooled estimates, because the endpoints are too heterogeneous to combine. No joint or cartilage endpoint appears anywhere in the 113 studies.
