Preclinicalcounts toward this tier
Supplying the precursor while blocking the drain: oral NMN plus apigenin in aged mice preserved NAD+, reduced cartilage-damage (OARSI) scores and hyaline-cartilage loss, and improved bone and muscle measures, with the mechanism mapped through the NAD+-dependent mitochondrial deacetylase SIRT3.
- Population
- Aged (20-month) mice and musculoskeletal precursor cells; SIRT3-knockout mice for mechanism
- Intervention
- Oral NMN plus the CD38 inhibitor apigenin (the 'N+A' regimen)
- Comparator
- Untreated aged mice
- Limitations
- About 5 animals per readout; not independently replicated; NMN-plus-apigenin is not the NMN-plus-resveratrol pairing consumers actually use. Full text read.
